Obtaining an interpretable and reliable biology result is the result of the mastery of the analytical technique, the sampling technique (type of tube, filling volume), the delay and the conditions of dispatch

Choose the right tube according to the examination to be made:
Tube colors are standardized and meet French, European and international standards.
Biochemical examination: use a dry tube (with or without gel separator) for a serum assay or a Heparin-Lithium tube for a whole blood or plasma assay.
Dosage on red blood cells (eg eGSH, eSOD): Imperative need for whole blood to isolate the red blood cell. The dry tubes (red) contain activators of coagulation, the globules thus form a clot which prevents us from isolating erythrocytes -> rejected tubes.

Choose the volume of tube adapted to the species and the examination to be carried out:
The amount of anticoagulant is calculated precisely according to the capacity of the tubes
(Eg: not reached filling level: dilution effect of the sample and disturbances of some analyzes due to excessive concentration of anticoagulant).

Fill the sample tubes:
Syringe sampling: Fast and gentle transfer of blood into the tube, taking care to remove the needle from the syringe to avoid iatrogenic hemolysis of the sample. Gently invert several times to homogenize the blood with the anticoagulant.

Recommended order if multiple tubes must be filled: Dry without additive, Citrate, Dry with additive, Heparin and EDTA. This minimizes coagulation interference.
Avoid interferences related to a bad choice of anticoagulant:
– Lithium heparin allows the realization of all biochemical analyzes, electrolytes and acid-base balance. Be careful, Sodium heparin obviously disturbs the Na + assays.
– Citrates also do not allow calcium assays, many enzymes, electrolytes and elements of acid-base balance.
– Fluoride tubes block glycolysis: imperatives for blood glucose determination.
– EDTA is a chelator; it traps metals -> impossible to measure Zn, Cu, Fe, Ca, Mg, … EDTA chelates calcium and as such is to be banned for biochemical assays in general. Indeed, a large number of biochemical assays use calcium as a cofactor and are therefore highly disturbed: Mg, creatinine, PAL, Amylase, for example, will be greatly underestimated. Conversely, interactions with electrolytic assays will yield highly overestimated K + and Na + results.

Lipemia, haemolysis and jaundice are three main pre-analytical conditions that may skew the result.

Lipemia (= lactescence)
It is caused by the presence of triglycerides. This is a change that is most often post-prandial and physiological (= non-pathological). A fasting of 12 hours makes it possible to avoid it. Some results may be biased (eg Na + and Cl- of the ionogram, measured by indirect potentiometry).

Most of the time, hemolysis is a severe in-vitro change, caused by a hard blood test / inappropriate needle diameter, serum (or plasma) separation time, freezing, or accidental overheating. whole blood, a delay in sending / receiving the sample and finally a lipemia (weakening of red blood cells).

There are several mechanisms that will skew certain analysis results during hemolysis:
– Changes in absorbance (eg creatinine, bilirubins, lipase, amylase, cholesterol, triglycerides) The degree of hemolysis required to alter the result may be variable.
– Interference and direct measurement of hemoglobin (eg total protein, ALT)
– Release of molecules of diagnostic interest or involved in the chemical reactions of the analyzer (eg potassium, folates, LDH, ASAT, iron and phosphorus)
– icterus

A jaundice sample often already reflects a pathological state and has as its main consequence an alteration of the absorbance which will distort the measurement of several analytes. This is especially true for creatinine, which can be lowered falsely.

Vitamins A, E and D3 and β-carotene degrade in the light.
-> protect the tube with aluminum foil for example.

Sample mixtures
Only a few methods have been validated to accept mixed samples.
Eg: NF U47 600-1 specifies that “the result obtained from a mixture of samples does not predict results that would be obtained from individual analyzes.”
The mixture of samples may cause a loss of sensitivity of the method by dilution, by reaction in the mixture produced, or by a phenomenon of camouflage related to the contacting of samples from animals not having the same status with respect to the disease sought.

Place the tube in the refrigerator before sending it to extend its conservation (+ 4 ° C maximum 2 days).


Sending preferably from Monday to Wednesday: avoid shipments on Friday (risk of damage during the weekend).


The use of outdated tube is not encouraged.

The tubes received before 11:00 am are treated the same day, the others can be postponed to the following day.


Primary tubes and secondary tubes after analysis are kept for two weeks. Then they are systematically destroyed. The sample remaining the property of the customer, if the latter wishes to recover it after analysis, he must make the request
